PLP-dependent Enzymes: a Powerful Tool for Metabolic Synthesis of Non-canonical Amino Acids

چکیده
An Overview on PLP-dependent Enzymes Pyridoxal 5’-phosphate (PLP), the biologically active of vitamin B6, was first identified in the mid-forties as the cofactor for the transamination reaction. Since then, PLP-dependent enzymes have been the focus of extensive biochemical research. The interest aroused by these enzymes is due to their unrivalled catalytic versatility and their widespread involvement in cellular metabolism. As a matter of fact, PLP acts as cofactor in more than 160 different enzymes classified by the Enzyme Commission, representing 4% of all known cellular catalytic activities [1]. PLP-dependent enzymes serve vital roles in all living organisms and catalyze a number of diverse chemical reactions, such as transamination, decarboxylation, racemization, carbon-carbon bond cleavage and formation. PLP-dependent activities are involved in essential biosynthetic pathways including glucose and lipid metabolism, 27

Molecular and phylogenetic analysis of pyridoxal phosphate-dependent acyltransferase of Exiguobacterium acetylicum.
The pyridoxal-5'-phosphate (PLP)-dependent family of enzymes is a very diverse group of proteins that metabolize small molecules like amino acids and sugars, and synthesize cofactors for other metabolic pathways through transamination, decarboxylation, racemization, and substitution reactions. In this study we employed degenerated primer-based PCR amplification, using genomic DNA isolated from ...
متن کاملDesign and Synthesis of New Benzimidazole and Pyrimidine Derivatives as α-glucosidase Inhibitor
In an endeavor to find a novel series of antihyperglycemic agents, new benzimidazole and pyrimidine derivatives were successfully synthesized efficiently in high yield with high purity, starting from amino acids in the presence of phosphorus oxychloride (POCl3). The synthesized compounds were identified by 1H-NMR, 13C-NMR, FT-IR spectroscopic techniques and elemental analysis. All products were...

متن کاملRidA Proteins Prevent Metabolic Damage Inflicted by PLP-Dependent Dehydratases in All Domains of Life
Pyridoxal 5'-phosphate (PLP) is a coenzyme synthesized by all forms of life. Relevant to the work reported here is the mechanism of the PLP-dependent threonine/serine dehydratases, which generate reactive enamine/imine intermediates that are converted to keto acids by members of the RidA family of enzymes. The RidA protein of Salmonella enterica serovar Typhimurium LT2 is the founding member of...

Background: Diabetes mellitus is one of the most common endocrine diseases. Cardiovascular disease (CVD) is one of the leading causes of death in patients with type 2 diabetes. The aim of this study was to investigate the metabolic profile of plasma amino acids in diabetic patients with cardiovascular disease. Methods: The present study is a descriptive-analytical cross-sectional study on 140 ...
